{"id":46,"date":"2007-04-19T13:58:41","date_gmt":"2007-04-19T20:58:41","guid":{"rendered":"http:\/\/www.bigroom.org\/wordpress\/?p=46"},"modified":"2007-04-19T13:58:41","modified_gmt":"2007-04-19T20:58:41","slug":"more-lambic-pictures","status":"publish","type":"post","link":"https:\/\/www.bigroom.org\/wordpress\/?p=46","title":{"rendered":"More Lambic pictures"},"content":{"rendered":"<p>Ah, that&#8217;s better &#8211; a more traditional heat-fix\/simple stain (using Methylene blue) shows my yeast isolates better:<\/p>\n<p><img decoding=\"async\" alt=\"Sally the maybe-Brettanomyces-type yeast\" title=\"Methylene Blue stained yeast, 1000X magnification\" src=\"http:\/\/www.bigroom.org\/images\/Sally_MB.jpg\" \/><br \/>\n(&#8220;Sally&#8221;, a yeast that I suspect is a Brettanomyces-type yeast.)<\/p>\n<p><img decoding=\"async\" alt=\"Sam the...Saccharomyces-type yeast?\" title=\"Methylene blue stain, 1000X magnification - small for a Saccharomyces?\" src=\"http:\/\/www.bigroom.org\/images\/Sam_MB.jpg\" \/><br \/>\n(&#8220;Sam&#8221; now looks awfully small&#8230;but more experienced observers than I am said that it could actually be a Saccharomyces-type yeast.)<\/p>\n<p><img decoding=\"async\" alt=\"Lucy the possibly-Pediococcus\" title=\"Lucy here may be another Pediococcus-type bacterium\" style=\"border: 2px none blue; float: left\" src=\"http:\/\/www.bigroom.org\/images\/LucyGram.jpg\" \/>I also got two more Coccoid-Cluster-type Gram-positive bacterial isolates.  The look pretty much the same under the microscope, though one had gooey wet, slightly larger colonies than the other&#8217;s smaller, hard-lump colonies.  I see another one of those <span class=\"moreinfo\" title=\"A group of 4\">tetrads<\/span> in the hard-lump-colony microscope image.<\/p>\n<p style=\"clear: both\">All told, I now have 10 isolates to check out.  I&#8217;ve been given the go-ahead to try sequencing on the 8 bacterial isolates so hopefully I&#8217;ll be able to get a clear identity for <span class=\"moreinfo\" title=\"I suspect Lactobacillus species\">Fred, Sid<\/span>, <span class=\"moreinfo\" title=\"I suspect Pediococcus-type species\">Lisa, Lucy, BillyBob, JimBob, BettySue, and MarySue<\/span>.
